Proteoform Thursday – Muhammad Zenaidee presents “Enhancing Structural Insights in Native Top-down Mass Spectrometry Experiments”

Structural biology aims to interrogate the three dimensional structures of proteins and their diverse proteoforms. Native top down mass spectrometry (TD MS), enhanced by ion mobility, enables detailed characterization of intact proteins, including sequence, post-translational modifications, and gas phase structure. Here, we introduce a custom software tool that improves interpretation of complex native TD MS data and enhances internal and terminal fragment assignments. Combining ion mobility with supplemental activation further reveals structural features such as disulfide bond connectivity, demonstrating that internal fragments can be utilized to significantly enhance structural analysis in native top-down mass spectrometry experiments.
