Proteoform Thursday: Christian Bleiholder presents “Top-Down Protein Analysis by Tandem-Trapped Ion Mobility Spectrometry/Tandem-MS”

We developed a novel platform combining collision-induced dissociation and UV photodissociation in a tandem-trapped ion mobility spectrometer coupled with subsequent automated MS/MS analysis. The instrumental setup allows (1) an MS3 workflow by combining two ion fragmentation modes and (2) the acquisition of cross-section information on the intact proteins and the fragment ions. This platform integrates top-down analysis of mobility-selected proteoforms with mobility-separation of fragment ions and their automated MS/MS characterization.
