Proteoform Thursday: Philipp Kaulich presents “Properties, Origin, and Reproducibility of Truncated Proteoforms in Recent Top-Down Proteomic Studies”

One of the most common modifications observed in top-down proteomics (TDP) studies is N-/C-terminal truncation, which can dramatically change biological function compared to the full-length proteoform. We performed a meta-analysis of more than 140,000 proteoforms reported by 50 different TDP data sets published over the past decade to examine the analytical and biological factors involved in termini and truncation site genesis, including the impacts of biological origin, sample preparation, data acquisition, and data analysis. In this talk, I will discuss the occurrence of artificially introduced truncated proteoforms, as well as the elucidation of potentially relevant proteoforms due to their presence in multiple independent TDP data sets.
