Proteoform Thursday: Jared Shaw presents “Ribosome Heterogeneity Revealed by Complex-Up Native Mass Spectrometry and Top-Down Proteomics”

We have developed a workflow combining complex-up native mass spectrometry with infrared multiphoton dissociation (IRMPD) and top-down proteomics to enable rapid and direct characterization of ribosome heterogeneity. IRMPD enabled preferential unfolding and fragmentation of 30S and 50S subunit rRNA for efficient ejection of the ribosomal proteins revealing heterogeneity in detail. TDP characterization of isolated ribosomal RPs enabled facile interpretation of the IRMPD spectra and provided insights into proteoform specific ribosome interactions.
