Proteoform Thursday: Oliver Hale presents “Enabling Molecular Pathology of Intact Proteoform Complexes in Tissue by Mass Spectrometry”

Proteoforms form complexes with other molecules including metal ions, small molecule cofactors and other proteoforms in order to function. Understanding the interactions of proteoforms complexes within the tissue environment is key to unravelling disease mechanisms. This presentation will detail advances made at the University of Birmingham that have enabled the analysis of soluble and membrane proteoform complexes directly from tissue. Using native top-down mass spectrometry and mass spectrometry imaging, complexes can be extensively characterised and spatially analysed. I will discuss the application of these technologies to understanding irregular binding of metal ions by the protein SOD1 in amyotrophic lateral sclerosis, a neurodegenerative disease.
